Without Charlie Sheen, the show is just pointless. He is the main character and the fact that his real life and his persona are almost the same people is pretty much why most people watch him. Replace Charlie with some other up and comer but I won't be watching and I know a lot of others won't be watching too. The show will not run without him.
As for Charlie Sheen himself, the dude has lost it. I think he has done so much drugs that it is starting to really fuck with this mental state. He can't think properly, contradicting himself all the time and just blabbering on about random bs. I will be surprised if the guy is still alive in the next 5 years.
